Purpose of the course This practical laboratory course is intended for PhD students who are interested in the cardiometabolic diseases and need to improve/expand their laboratory skills. The course aims to teach the PhD students the basics of commonly used laboratory techniques used to study inflammation, atherosclerosis, insulin resistance, obesity and other pathological conditions related to cardiometabolic diseases. The students will obtain a basic understanding of several commonly used laboratory techniques, learn the theory behind them, how to avoid common mistakes and pitfalls and get a chance to view and/or perform many of the techniques themselves.
Intended learning outcomes At the end of the course, the student is supposed to be able to:
• Name and describe basic research methods that are commonly used to study cardiometabolic diseases.
• Justify the use of a certain method and what results can be obtained using it
• Describe how to perform the methods using standard protocols
• Apply these methods to their own research question and material
• Identify common pitfalls and how to avoid them
Contents of the course The research methods taught in the course will include mouse dissection demonstration, cell preparation of lymphoid organs and subsequent analysis by flow cytometry or sorting for bulk mRNA-seq or scRNA-seq, isolation of the heart, liver and major arteries, preparation and analysis of histological sections for confocal microscopy or pathology, isolation of adipose tissue and spheroid culturing and analysis, RNA extraction and subsequent steps for mRNA-seq. Note that this is not an animal experimentation course and therefore the course does not certify students to dissect animals themselves, only gives them a demonstration to it.
Teaching and learning activities The course is a practical course with laboratory demonstrations and a possibility for all students to enhance their skills.
Compulsory elements Presence at all practical demonstrations and lectures. If absent the student can compensate by performing a written assignment about the part that was missed.
Examination Group assignment with oral presentation at the end of the course to show understanding of the demonstrated techniques, when to use them and how to perform them correctly.
Literature and other teaching material Protocols for each method being taught will be provided at the start of the course.

Selection of students Selection will be based on 1) the relevance of the course syllabus for the applicant's doctoral project (according to written motivation), 2) start date of doctoral studies (priority given to earlier start date)
